 EXPERIMENT #1 QUESTION: DOES LACTAID© DIGEST LACTOSE AS ADVERTISED?Aspergillus oryzae Some lactose intolerant individuals take LACTAID before consuming milk products. LACTAID pills contain LACTASE enzyme isolated from the fungus as opposed to human lactase.Producers of LACTAID claim the pills reduce the adverse symptoms typically experienced by the lactose intolerant.Experiment #1 Hypothesis:1.     LACTAID lactase WILL / WILL NOT (circle one) digest the lactose sugar found in milk. Explain your reasoning.   Experiment #1 Setup:1.     Use your permanent marker to label 5 separate 100 mL beakers with the following: Lactaid (lactase), Milk (lactose), Table sugar (sucrose), Denatured Lactaid, and Water.2.     Prepare solutions as described in a-e. Use a wooden stick to mix each solution, taking care that each substance is thoroughly dissolved in solution.a.      Use the mortar and pestle to crush the lactaid tablet. Dissolve the powder in 100 ml of tap water in the Lactaid (lactase) beaker.b.     Dissolve 5 grams of dried milk in 50 ml of tap water in the Milk (lactose) beaker.c.      Dissolve 5 grams of sugar in 50 ml of tap water in the Table sugar (sucrose) beaker.d.     Pour 50 ml of solution from Lactaid (lactase) cup into the Denatured Lactaid beaker. Add 5 grams of baking soda to this beaker. Stir to dissolve.e.      Fill with 100 ml of plain tap water.3.     Using forceps, select a glucose strip and dip the blue-colored end into the Lactaid (lactase) solution. Remove the strip immediately and then wait 30 seconds to compare the strip’s color to the color key illustrated below. Light blue color indicated glucose is absent. Anything other than the light blue color indicates that glucose is present. Strip color varies from green to brown depending on glucose concentration.  Record whether glucose is present/absent in the table below. Repeat this process for the rest of your solutions, using a new glucose strip each time. Lactaid (lactase) Absent milk (lactose) Absent table sugar (sucrose) Absent Denatured lactaid (baking soda) Absent tap water Absent After the testing these are the above results    Experiment #1 Procedure:1.     Use permanent marker to label one test tube with “A-1” and another test tube “B-1”.2.     Use a pipette to add 3 ml of milk (lactose) to test tubes A-1 and B-1. Each hashmark on the pipette is equal to 0.5 ml, so filling the pipette to the line just below the bulb is equal to 3ml. Keep that pipette in your milk solution3.     Use a DIFFERENT pipette to add 3 ml of LACTAID (lactase) solution to test tube A-1 and wait 2 minutes. Keep that pipette in your Lactaid solution.4.     After the 2-minute wait, use forceps to dip a glucose strip into the solution. Remove the strip immediately and then wait 30 seconds to compare the strip’s color to the color key illustrated on previous page. Record whether glucose was present or absent in the Results Table 1 below.5.     Use a new pipette to add 3 ml of tap water to test tube B-1 and wait 2 minutes. Keep that pipette for you water solution. Using forceps, dip a new glucose strip to repeat step #4 for tube B-1.   6.     DO NOT throw away any solutions! You will use them in Experiment #2 and #3.7.     Empty contents of test tubes A-1 and B-1 down sink with water, carefully rinse with fresh water, return to test tube rack upside down to facilitate drying.Experiment #1 Results Table: Test Tube Solution Glucose present or absent?
